Abbreviations
(i) Genes: EYFP - enhanced yellow fluorescent protein; ECFP - enhanced cyan fluorescent protein; HN - heamagluttinin/neuraminidase gene from virus SV5 (type II signal-anchor sequence); pac - puromycin resistance
2A - self-cleaving 23aa sequence, 2Amut - inactive, point mutant of 2A.
(ii) Targeting Signals: MT - mitochondrial targeting sequence; NC - nuclear localisation signal; GT - GalT Golgi targeting (type II signal-anchor sequence); CR - calreticulin endoplasmic reticulum (ER) targeting type I (cleaved) signal sequence.
The constucts shown below generate multiple proteins from a single gene.
Cells are transfected with a single plasmid.
Cell-lines expressing all of the proteins can be rapidly and efficiently established by puromycin selection - resistance being conferred by the pac gene.
